This number of indentations is required for the final calculation of the modulus of elasticity. The glass is activated by applying various coatings described in the protocol. Due to the relative size of the AFM scan head, it is not possible to use a petri dish to store the polyacrylamide gel during the experiment. The active cover has a diameter of about three centimeters and serves as a mounting platform for samples. Unlike a petri dish, these sliders are smooth and do not interfere with the wet room or the AFM scan head. The caps themselves have yet to be activated, which is related to steps one to six of Dr. Wang’s protocol.

After determining the specific polyacrylamide concentration, about 200 ml is applied to the activated cover glass using a transfer pipette  , and a smaller round or square cap is placed  over the gel.
